چکیده
ABSTRACT Zinc is employed as a supplement; however, zinc-related nephropathy is not generally known. In this study, we investigatecl zinc-induced renal cell iajury using a pig kidney-derived cultured renal epithelial cell line, LLC-PKI, with proximal kidney tubule-like features, and examined the involvement of free radicals and extracellular signal-regu]ated kinase (ERK) in the cell injury. The LLC-PKI cells showed early uptake of zinc (30 pM), and the release of lactate dehydrogenase (LDH), an index of cell iajury, was observed 24 hr after uptake. Three hours after zinc exposure, generation of reactive oxygen species (ROS) was increased. An antioxidant, IV] N'-diphenyl-p-phenylenedi-

Aldosterone Induces Oxidative Stress Via NADPH Oxidase and Downregulates the Endothelial NO Synthesase in Human Endothelial Cells
Aldosterone is traditionally viewed as a hormone regulating electrolyte and blood pressure homeostasis. Recent studies suggest that Aldo can cause microvascular damage, oxidative stress and endothelial dysfunction. However, its exact cellular mechanisms remain obscure. This study was undertaken to examine the effect of Aldo on superoxide production in human umbilical artery endothelial cel...
متن کاملSpironolactone Inhibits NADPH Oxidase-Mediated Oxidative Stress and Dysregulation of the Endothelial NO Synthase in Human Endothelial Cells
Accumulating evidence indicates that aldosterone plays a critical role in the mediation of oxidative stress and vascular damage. NADPH oxidase has been recognized as a major source of oxidative stress in vasculature. However, the relation between NADPH oxidase in aldosterone-mediated oxidative stress in endothelial cells remains to be ascertained. The present study aimed to investigate the rel...

متن کاملPhosphatidic acid and diacylglycerol directly activate NADPH oxidase by interacting with enzyme components.
The enzyme NADPH oxidase is regulated by phospholipase D in intact neutrophils and is activated by phosphatidic acid (PA) plus diacylglycerol (DG) in cell-free systems. We showed previously that cell-free NADPH oxidase activation by these lipids involves both protein kinase-dependent and -independent pathways. Here we demonstrate that only the protein kinase-independent pathway is operative in ...
متن کاملRole of alpha1-adrenoreceptors in cocaine-induced NADPH oxidase expression and cardiac dysfunction.
OBJECTIVE We assessed whether alpha1-adrenoreceptor (alpha1-AR) stimulation contributes to activation of myocardial NADPH oxidase in a rat model of cocaine-induced cardiac dysfunction. METHODS AND RESULTS After 7 days of cocaine injection (2 x 7.5 mg/kg/day, i.p., Coc), NADPH activity assessed by chemiluminescence increases as well as phosphorylation of p47phox, one of the cytosolic component...
متن کاملCritical role of microglial NADPH oxidase-derived free radicals in the in vitro MPTP model of Parkinson's disease.
1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) damages dopaminergic neurons as seen in Parkinson's disease. Although increasing evidence suggests an involvement of glia in MPTP neurotoxicity, the nature of this involvement remains unclear. Exploiting the advantage of cell culture systems, we demonstrated that microglia, but not astroglia, significantly enhanced the progression of MPTP-indu...
